This application note demonstrates the superior quality and editing performance of Synthego’s chemically modified sgRNAs relative to modified sgRNAs from Vendor D.

Synthetic sgRNAs offer an unbeatable combination of quality, speed, accuracy, and price for CRISPR experiments. Another advantage of sgRNAs is that they can also be chemically modified to protect them from intracellular immune responses and RNA-degrading exonucleases. Such modifications are especially valuable when editing challenging cell types such as primary cells and stem cells.

Although chemically modified synthetic sgRNAs (mod sgRNAs) are now available from several vendors, they are not all equal in quality. The purity of sgRNAs, measured by the percentage of full-length product (the active portion of the total RNA), may vary widely across manufacturers. As impure sgRNAs may result in higher off-target binding or non-binding events, high quality sgRNA is critical to achieving high editing efficiency in CRISPR-Cas9 experiments.

Synthego’s chemically modified synthetic sgRNAs outperform sgRNA from Vendor D in many areas, including:

  • Having higher sgRNA quality and purity
  • Exhibiting higher editing efficiencies
  • Creating gene knockouts with higher frequency in primary T cells
