This application note demonstrates the high editing performance of Synthego’s chemically modified sgRNAs without HPLC purification. The results presented here indicate that our high-quality synthesis removes the need for an HPLC purification step required by other vendors, decreasing both turnaround time and cost of Synthego’s sgRNAs.
Synthetic sgRNAs offer an unbeatable combination of quality, speed, accuracy, and price for CRISPR experiments. Synthetic sgRNAs can also be chemically modified to protect them from degradations and prevent triggering intracellular immune responses. These modifications are especially valuable when editing challenging cell types, such as primary cells and stem cells.
Synthetic sgRNA products from other vendors contain impurities that can increase off-target binding and reduce on-target binding. Although many vendors rely on extra HPLC purification to provide high-quality sgRNAs, this extra step adds time and cost to the order. Synthego produces exceptionally pure products through our standard manufacturing process, without a requirement for HPLC purification.
Synthego (non-HPLC) chemically modified sgRNAs and HPLC-purified sgRNAs produced internally showed comparable editing efficiencies. Eliminating the need for HPLC purification saves researchers time and money.
